【Principles】
Vitamin B9 (Folic Acid) Test Kit used microbiology method to detect the volume of Folic Acid in food matrices. Folic Acid supplies the growth of Lacticaseibacillus rhamnosus. Its growth rate associated with the volume of Folic Acid within the sample. Take the Folic Acid medium (Lacticaseibacillus rhamnosus added), samples or Standards and add to the microwell. Incubate at 37℃ and avoid light exposure. Lacticaseibacillus rhamnosus will start growing until Folic Acid are consumed entirely. It takes 44-48 hours to complete incubation. Use microplate reader to read the sample turbidity with 610-630nm or 540-550nm filter. The volume of Folic Acid in sample can be determined by comparing the sample turbidity with the Standards.

—- Phosphate buffer (0.05mol/L, 0.1% Ascorbate, pH 7.2): Add 7.8g NaH2PO4·H2O (Sodium dihydrogen phosphate dihydrate) and 1g Sodium ascorbate to 1L redistilled water and dissolve completely, adjust pH to 7.2. Prepare it fresh before each use.
