Alkaline Phosphatase Rapid Test Kit

Alkaline Phosphatase (ALP) is a heat sensitive enzyme that is naturally present in raw milk, and it can be destroyed by conventional pasteurization. This kit can determine whether the samples meet the U.S. and E.U. pasteurization requirements of by determining the content of ALP.

ALP activity (mU/L) can be detected by alkaline phosphatase module in PureTrustTM Intelligent Fluorescence Detector. The sample is positive when the detection value is greater than or equal to 350mU/L. The sample is suspicious when the detection value is 100~350mU/L. The sample is negative when the detection value is less than 100mU/L.

High-temperature short-time sterilized milk or stable sterilized milk at room temperature may contain reactivated alkaline phosphatase, so they cannot be detected by this reagent.

Assay Principles

The kit uses chemiluminescent substrate of specific enzyme to determine the activity of alkaline phosphatase in dairy products, which can quickly verify the pasteurization effect within 2 minutes. The chemiluminescent substrate can release photons under the catalysis of ALP. Within a specific concentration range of ALP, the number of released photons has a good linear relationship with ALP concentration in the sample. Combined with PureTrustTM Intelligent Fluorescence Detector, the operator can quantitatively detect ALP concentration in samples. Before testing samples, negative and positive control need to be used to calibrate and establish Pass/Fail RLU limits.

Sensitivity

Detection limit in pasteurized milk (75℃, 15 seconds) is 20mU/L (approximately 0.002% raw milk).

With the presence of hydrogen peroxide, it is capable of oxidizing thiocyanate ions to produce products with broad antimicrobial activity. Lactoperoxidase has an important role in the dairy cattle immune system. Lactoperoxidase is one of the most heat-stable enzymes found in milk. Pasteurization cannot deactivate it completely. Ultra-high-temperature processing will deactivate it completely.
